Purpose: To compare valacyclovir to acyclovir in preventing recurrences of herpes simplex virus (HSV) eye disease. Methods: Fifty-two patients with recurrent HSV disease were randomized (25 with valacyclovir 500 mg/day; 27 with acyclovir 800 mg/day for one year). Results: Recurrence rate of ocular HSV disease was 24.0% in the valacyclovir group, 22.2% in the acyclovir group (P > .05). No differences were found in the two groups regarding adverse reaction (most frequent were nausea, headache). No serious complications occurred in either group. Conclusion: In this study, the efficacy of onr-year suppression therapy with oral valacyclovir was comparable to acyclovir in reducing the recurrence rate of ocular HSV.
