Super-resolution microscopy using photoswitchable molecules requires fast and efficient software for single molecule detection, localization, drift correction, and visualization. However, there is still a dearth of freely available, flexible and easy-to-use reconstruction software coupled with image acquisition. Here, we present QuickPALM, an ImageJ plugin enabling real-time reconstruction of 3D super-resolution images during acquisition and drift correction. We illustrate its application by reconstructing Cy5-labelled microtubules with a resolution of 40-50 nm.
